Moving from standard CAR T-cells to TriCAR T-cells represents the next frontier in cellular immunotherapy designed to overcome antigen escape-driven relapse. Duo TriCAR enables expression of two CAR molecules on each LVV-transduced T cell — targeting three cancer antigens simultaneously.
Targets (B-Cell Malignancies)
Drastically Lowers Relapse Rates. By shutting down the primary avenue of tumor resistance (antigen escape), remissions are expected to be far more durable and permanent.
Overcomes Tumor Heterogeneity. A TriCAR T-cell can wipe out the entire diverse population of cancer cells, rather than just a CD19-positive subset.
Expanded Patient Eligibility. Patients whose leukemia naturally lacks CD19 can still be treated if they express CD20 or CD22 — a group excluded from standard CAR T therapy.
Triple Anti-Tumor Redundancy. For the cancer to escape, it would need to simultaneously mutate and lose all three antigens — a biologically rare event.
A single engineered T-cell equipped with two different chimeric antigen receptors — one targeting mature plasma cells (BCMA) and one targeting early B-cell precursors (CD19). One cell, two bullets. This "one product, multiple usage" platform offers unprecedented versatility.
Targets
Prevents Antigen Escape in Multiple Myeloma. Anti-BCMA eliminates the mature tumor mass while anti-CD19 hunts down the myeloma stem cells that cause relapse.
Immune Reset for Autoimmune Diseases. Completely purges the entire lineage of antibody-producing cells, effectively "rebooting" a patient's immune system to a healthy state in SLE and Systemic Sclerosis.
Organ Transplant Desensitization. Clinical trials show successful desensitization of "highly sensitized" patients, enabling compatible kidney transplants after years on waiting lists.
One Product, Multiple Indications. A single manufacturing platform addressing oncology, autoimmunity, and transplant medicine — expanding market access and clinical utility.
A blood-based immunotherapy (also called photochemotherapy) where white blood cells are separated, treated with psoralen (8-MOP), exposed to UVA light (365 nm, 2 J/cm²), and safely returned to the patient. This process reprograms T-cells to either suppress unwanted immune attacks or stimulate targeted cancer destruction.
Mechanism
CTCL — First-line FDA-approved therapy for advanced Sézary syndrome and mycosis fungoides. Overall response rates ~43%, complete response in 10% of patients.
GVHD — Second-line for steroid-refractory cases. Highly effective in both acute and chronic GVHD, particularly when initiated early after diagnosis.
Organ Transplant Management. Used in heart and lung transplants to reduce immune rejection while allowing physicians to reduce toxic immunosuppressive medication loads.
Emerging Autoimmune Applications. Off-label exploration in Crohn's disease, systemic sclerosis, atopic dermatitis, and other refractory immune conditions.
